Is the ink water based?

Yes it is waterbased. It is my go-to pen for detail on painting rocks and such. I love this pen. I bought several of them in all the sizes it comes in. MY FAVORITE!!!!! You can do a light coat of spray clear over it also and it wont bleed like sharpie pens do when you clear over them. If you know anybody that paints rocks this is a MUST have!!!!!

Will this pen give me fine upstroke and thicker downstrokes to mimic a calligraphy font when writing in small size lettering?

Yes, it will. It may be difficult for extremely small lettering, but the flexible point does allow you to mimic calligraphy.
