What are eyes made of and how are they attached?

The eyes, nose and mouth are embroidered to the face. They won't come out.

Do the pants come off?

Yes - it is actually overalls and they do come off. A great toy that is fun just as a toy and with lots to learn with all the colors and things a child can do.
