What is the holes spacing on the 8 x 4 3-way vent?

The screw holes are 12 and 3/4 inches...center to center. It fit perfectly the prior vent cover.

What is the distance between screw holes?

10 and 3/4 (10.75) inches. I replaced the original vent cover and it fit fine.

will a 4 x 10 fit in a 3 3/4 x 9 1/2 opening?

Yes it will work because the iner dimensions is smaller then the outside diameter so your good.if u have a hole exactly the sides of this vent then it won't cuz it won't have to were to screw too.so yes your good this is perfect fit for the measurements your have...

I ended up returning because it was thin and cheap. I went to home depot and bought a metal one for cheaper and painted it. Hope this helped. Michelle
