What is the thread measurement on the end of the spoke?

It's actually 2-56. Thats a #2 size with 56 threads per inch. The spokes will come with the threaded sleeves.

Can the spoke screw through the end of the nipple if needed?

There is about 10mm of threads on the spoke.. when the threads run out, the spoke can not be threaded any further. the distance the spoke can "thread through' the nipple is governed by the Depth of the threading inside the nipple. a 12mm nipple will have about 8mm of thread inside... a 14mm nipple has slightly more, the 16mm has more threads.... and the Amounts vary from brand to brand... a SHORTER nipple will allow more "thread-through" than a longer nipple. Your best choice is to error to the Short Side on spoke length. If a calculator says 257, get a 256, etc... If you try to go PAST the binding point, you damage the nipple and spoke... the nipple takes most of the damage.

What size spokes do I need for a 20”fat tire?

I was replacing the old spokes because 4 were broken. I looked online for how to measure spokes, then I measured one of the unbroken spokes. Note that the front wheel has one length. The back wheel has two sizes. The drive side is shorter. So there are three sizes. I used a carpenters tape to measure the old spokes. The new ones were just right.
