Can I use this without a caulking gun?

The quick answer is yes, and you don't have to scoop it out with your hands. But the truth is that it is much easier with a caulking gun and that is what it is made for. You can cut the tip off and push from the bottom. You would not have good control and the more you use the harder it gets. Spend $9 and get a caulking gun.

How hard is this when it dries? Would it make a solid "shim" if injected into a 1/8 to 1/4 gap?

Even if it did get that hard I would not do it. It is not what it was made to do.why would you not use wood shims?

would this be good for adhering a single large 6"x24" back splash tile to painted drywall

Heavy Duty Construction adhesive will work at attaching the tile to drywall. Make sure the paint coating is in good condition to ensure a proper bond between the tile and drywall. Also, make sure the adhesive and substrates are above 60* F for the best bond.
